Testing the Drug Psilocybin in Adults with Treatment-Resistant Depression

Treatment-resistant depression (TRD) is when a person receives help for depression such as therapy with a physiatrist and taking an anti-depression drug, but depression keeps coming back. This study will test a drug called Psilocybin, to treat patients with this condition. The study wants to see if the drug will better help people with this disease and is safe. People in this study will be randomly chosen to receive psilocybin or a placebo. The drug and placebo are taken as a swallowed pill. The placebo is a pill without the treatment. Being in the study requires participation for 55 to 62 weeks. Participants will attend clinic visits in person at a University of Utah clinic. Medical tests will be done during the study to track the health of participants.

Inclusion Criteria
- Ages18 years and older
- Diagnosis of Major Depressive Disorder
- Failure to respond to 2-4 different antidepressant medications for the current depressive episode
Exclusion Criteria
- Bipolar disorder
- History of psychotic disorder (schizophrenia, schizoaffective disorder)
- Alcohol or substance use disorder over the last 12 months
- Pregnancy or breastfeeding
- History of stroke, heart attack, seizure, or uncontrolled high blood pressure.
